Most of the functional properties of glycoprotein are dependent on the glycosylation pattern and occurrence of glycosylation at the probable sites. The N-glycosylation profile of the glycoprotein can be generated by cleaving the Asp-linked N-glycan from protein by PNGase F and analyzed by HILIC–MS technique. The linkage of each glycan form can be confirmed through the digestion with specific exoglycosidases and the confirmed through HILIC-MS and MS/MS. The column and system are equilibrated with dextran ladder before each set of HPLC runs which also provide the GU value of each glycoforms and can act as a standalone analysis technique.
